چکیده

Beckman et al. (1) reported altered monoaminergic systems and depressive-like behaviors in a line of mice lacking the cellular prion protein PrP (congenic B10.129-Prnp mice). These mice were derived from gene-targeted 129/Ola embryonic stem cells and were subsequently crossed to C57BL/10SnJ mice. Consequently, the genomic region of B10.129-Prnp mice flanking the targeted Prnp locus contains a stretch of up to 47.4Mb isogenic to 129/Ola (2). In this region, any genes polymorphic between the 129/Ola and C57BL/10SnJ strains will display 129/Ola allelotypes (2). This genetic anomaly represents a systematic confounder when congenic knockout mice are compared with wild-type mice of the back-crossing strain, even when using littermates from heterozygous breedings (2). This phenomenon, intrinsic to all congenic knock-outmice, is known as the flanking gene problem.

Hydroethanolic extract of Carthamus tinctorius induces antidepressant-like effects: modulation by dopaminergic and serotonergic systems in tail suspension test in mice

Objective(s): Studies indicate that major deficiency in the levels of monoaminergic transmitters is a reason for severe depression. On the other hand, it is shown that Carthamus tinctorius L. (CT) may improve neuropsychological injuries by regulation of the monoamine transporter action. Hence, the present study was undertaken to evaluate the involvement of monoaminergic systems in antidepressan...

O-10: Formation and Molecular Composition of The Sperm Head to Tail Coupling Apparatus

Background According to a worldwide survey in 2010 infertility affects 48.5 million of couples. In roughly half of the cases infertility is provoked by the male mate. Thus, a significant percentage of young men are infertile but the underlying causes are mostly unknown. Male fertility and reproduction success critically depends on proper formation of the mature sperm. Transmission of the geneti...

Rines E3 ubiquitin ligase regulates MAO-A levels and emotional responses.

Monoamine oxidase A (MAO-A), the catabolic enzyme of norepinephrine and serotonin, plays a critical role in emotional and social behavior. However, the control and impact of endogenous MAO-A levels in the brain remains unknown. Here we show that the RING finger-type E3 ubiquitin ligase Rines/RNF180 regulates brain MAO-A subset, monoamine levels, and emotional behavior. Rines interacted with MAO...

Altered neuron excitability and synaptic plasticity in the cerebellar granular layer of juvenile prion protein knock-out mice with impaired motor control.

Although the role of abnormal prion protein (PrP) conformation in generating infectious brain diseases (transmissible spongiform encephalopathy) has been recognized, the function of PrP in the normal brain remains mostly unknown. In this investigation, we considered the effect of PrP gene knock-out (PrP(0/0)) on cerebellar neural circuits and in particular on granule cells, which show intense P...

Nanocurcumine Ameliorates Lipopolysaccharide-induced Depressive-like Behavior in Mice

Objective(s): Curcumin, a plant alkaloid from Curcuma longa, possess antioxidant and anti-inflammatory properties. Recently, the antidepressant activities of curcumin were reported. Nevertheless, bioavailability of curcumin limits its therapeutic utility. Nanotechnology is a developing field that potentially enhances bioavailability and the plasma concentration of curcumin. This study investiga...
